概要 | Many central neurons support active dendritic spikebackpropagation mediated by voltage-gated currents. Active spikesin dendrites have been shown capable of providing feedback to thesoma to influence somatic excitability and firing dynamicsthrough a depolarizing afterpotential (DAP). In pyramidal cellsof the electrosensory lobe of weakly electric fish, Na(+) spikesin dendrites undergo a frequency-dependent broadening thatenhances the DAP to increase somatic firing frequency. We use acombination of dynamical analysis and electrophysiologicalrecordings to demonstrate that spike broadening in dendrites isprimarily caused by a cumulative inactivation of dendritic Na(+)current. We further show that a reduction in dendritic Na(+)current increases excitability by decreasing the interspikeinterval and promoting burst firing. This process arises wheninactivation of dendritic Na(+) current shifts the latency of thedendritic spike to delay the arrival of the DAP sufficiently toincrease its impact on somatic membrane potential despite areduction in dendritic excitability. Furthermore, therelationship between dendritic Na(+) current density and somaticexcitability is nonmonotonic, as intermediate levels of dendriticNa(+) current exert the greatest excitatory influence. Theseresults reveal that temporal shifts in dendritic spike firingprovide a novel means for backpropagating spikes to influence thefinal output of a cell. After the completing the simulation, Fig.2B in Fernandez et al, JNP2005 is shown up. |
